Some of the key steps in the synthesis of this molecule involve: 1. Formation of the seven-membered azepane motif through an azide-olefin cycloaddition. 2. Preparation of the quaternary center via an aza-Cope/Mannich rearrangement from a bicyclic alcohol. 3. A diastereoselective 1,2-addition of a vinyl species to a ketone to generate the bicyclic alcohol. 4. Formation of a bicyclic ketone from a desymmetrization and a Beckmann rearrangement of a symmetrical bicyclic diketone. I set up a Horner–Wadsworth–Emmons reaction. This reaction allows the stereoselective olefination of aldehydes or ketones using stabilized phosphonate carbanions. Phosphonate carbanions are more nucleophilic than the corresponding phosphorus ylides, allowing them to react with a wide range of aldehydes and ketones under milder conditions. Hindered ketones that are typically nonreactive in Wittig reactions can react in Horner–Wadsworth–Emmons olefinations. The reaction favors the formation of (E)-alkenes. When electron-withdrawing phosphonates are employed, the elimination of the oxaphosphetane intermediates accelerates, boosting the production of (Z)-alkenes. This is known as the Still–Gennari modification. In addition to the type of phosphonate used, the selectivity of the reaction can also be influenced by other factors such as the choice of base and solvent. The Knoevenagel condensation enables the reaction of aldehydes or ketones with active methylene compounds (containing two electron-withdrawing groups, such as malonic esters, acetoacetic esters, acetylacetone, or cyanoacetic acid) in the presence of a weak base to yield α,β-unsaturated dicarbonyl or related compounds. When the reaction is carried out with carboxylic acids, such as malonic acid, in the presence of pyridine as the solvent, the condensation is followed by a pyridine-induced decarboxylation. One of the most convenient ways to perform this reaction is by using the Ohira–Bestmann reagent. Deacylation of the Ohira–Bestmann reagent by a base, such as potassium carbonate, generates a carbanion, which reacts with the carbonyl group of an aldehyde.
